The pair has starred in some of Bollywood's biggest hits such as Dilwale Dulhania Le Jayenge, Kuch Kuch Hota Hai and Kabhi Khushi Kabhie Gham.
Their new film, called My Name is Khan, is aiming for a 2009 release, director Karan Johar said during the weekend.
Johar declined to reveal the plot but some newspapers say Khan will play a Muslim man and highlight the plight of the community following the attacks in the US on September 11, 2001.
